It’s only one little spot. Could it be a disease or i also have a plant in there, could it be irritating him?
It sounds as if your Betta has rubbed against something. It could also make him prone to disease. Perhaps raise the temp to about 78 degrees and add 1 tablespoon of aquarium salt per 5 gallons of water to your tank as a precaution(dont pour it straight in, let it disolve thru a net or something).

My goldfish used to die from getting scratched up by plants I had in their bowl, but my Betas never did. Maybe take the plants out of the tank and see if he loses more scales.

It’s unlikely that the plant is irritating your fish. Is it possible that he scraped up against something hard in the tank?
Another possiblity is some sort of skin ulcer or parasite. Is he otherwise acting normally?
